Vehicles in Mafia: Definitive Edition consist of over 50 passenger cars, trucks, and motorcycles that are all fitting to the setting of the game.

- Free Ride vehicles are unlocked in Free Ride mode.
- Downloadable content (DLC) vehicles are obtained through the Chicago Outfit Pack.
- Game Ownership Bonus (GOB) vehicles are obtained if player owns Mafia III: Definitive Edition, Mafia II: Definitive Edition, or Mafia: Trilogy.
- My2K bonus vehicles are obtained by linking your game to a valid My2K account.
